General Information:

Risperidone is an atypical antipsychotic medication used to treat schizophrenia, bipolar disorder, and irritability associated with autism spectrum disorder (ASD). It works by modulating the activity of dopamine (D) and serotonin (5-HTA) receptors in the brain, helping to control mood, thought processes, and behavior.

Solubility: Slightly soluble in water, freely soluble in methanol and acetic acid

Mechanism of Action:

Blocks dopamine D receptors, reducing symptoms of psychosis and hallucinations.

Blocks serotonin 5-HTA receptors, improving mood and cognitive symptoms.

Has mild antagonistic effects on histamine (H) and alpha-adrenergic (, ) receptors, which can cause sedation and dizziness.

Absorption: Well absorbed orally, peak plasma levels in 12 hours.

Bioavailability: 70% (oral administration).

Metabolism: Primarily in the liver (via CYP2D6) into active metabolite 9-hydroxy-risperidone.

Half-life:Risperidone: 36 hours

Active metabolite (9-hydroxy-risperidone): 2024 hours

Excretion: 70% via urine, 14% via feces. Therapeutic Uses & Indications:

Schizophrenia (adults and adolescents).

Bipolar disorder (manic episodes). Irritability and aggression in autism spectrum disorder (ASD). Off-label uses:

Treatment-resistant obsessive-compulsive disorder (OCD).

Post-traumatic stress disorder (PTSD).

Severe Tourette syndrome.

Dosage & Administration:

Schizophrenia:

Adults: 2 mg/day, increasing up to 46 mg/day if needed.

Elderly: Start at 0.5 mg twice daily.

Bipolar Disorder:

Adults: 13 mg/day, up to 6 mg/day.

Autism Spectrum Disorder (ASD) (Children 516 years):

Starting dose: 0.250.5 mg/day, increased based on response.

Depot Injection (Long-Acting Form Risperdal Consta):

IM Injection every 2 weeks (2550 mg).
